Based on the fact that bisulfite can deamination cytosine without methylation in DNA and convert it into uracil, while the methylated cytosine remains unchanged, PCR amplification of required fragments will convert all uracil into thymine, and then high-throughput sequencing of PCR products will be carried out. By analyzing the methylation difference between different samples, we can study the regulation of DNA methylation level on gene expression.
